Abstract Wheat is the most widely cultivated food crop on the planet, and it feeds the majority of the world's population. The field experiment was performed from 2019 to 2021 in the eastern part of Loess Plateau, China Nitrogen (N) fertilizer application improves winter wheat yield. It was necessary to analyze the nitrogen utilization of different varieties and screen varieties with high yield and high nitrogen utilization. Six dryland wheat varieties YH-20410, YH-618, YH-805, JM-92, C-6359, and LH-6 were selected to compare the differences in nitrogen uptake and utilization yield, and quality. The aim was to provide theoretical basis for high yield, and high-efficiency wheat production in dryland of southeast loess Plateau. The results showed that the rainfall during the fallow period from 2019 to 2021 was 245.5 mm and 288.9 mm, respectively, and the nitrogen application rate was 180 kg ha-1.The highest transshipment amount, and transshipment rate of nitrogen before anther were found in YH-618, which reached 79.4 kg ha-1 and 58.6%, respectively. The yield of wheat cultivar YH-20410 was the highest, reaching 4890 kg ha-1, which was significantly higher than that of YH-805, JM-92, C-6359 and LH-6, but there was no significant difference of YH-618. The grain protein content of YH-618 was the highest (15.2%), which was not significantly different from that of JM-92, YH-805 and LH-6, but significantly higher than that of YH-20410, and C-6359, respectively. Therefore, in high-yield years, fallow cultivation can help adjust the relationship among the components, promote a reasonable distribution, and improve yield.

ABSTRACT Purpose: To investigate the effects of Periplaneta americana L. on ulcerative colitis (UC) induced by a combination of chronic stress (CS) and 2,4,6-trinitrobenzene sulfonic acid enema (TNBS) in rats. Methods: The experiment UC model with CS was established in rats by a combination of chronic restraint stress, excess failure, improper, and TNBS. The body weight, disease activity index (DAI), colonic mucosal injury index (CMDI), histopathological score (HS) and pro-inflammatory mediators were measured. The content of corticotropin-releasing hormone (CRH) in hypothalamus or adrenocorticotropic hormone (ACTH) and corticosteroids (CORT) in plasma were evaluated by enzyme-linked immunosorbent assay. The proportion of T lymphocyte subsets was detected by flow cytometry, and gut microbiota was detected by 16S rDNA amplicon sequencing. Results: Weight loss, DAI, CMDI, HS and proinflammatory mediators were reversed in rats by P. americana L. treatment after UC with CS. Increased epidermal growth factor (EGF) was observed in P. americana L. groups. In addition, P. americana L. could reduce the content of CRH and ACTH and regulate the ratio of CD3+, CD3+CD8+ and CD3+CD4+CD25+/CD4+ in spleen. Comparably, P. americana L. changes composition of gut microbiota. Conclusions: The ethanol extract of Periplaneta Americana L. improves UC induced by a combination of CS and TNBS in rats.

A cDNA encoding chalcone synthase (CHS), the key enzyme in flavonoid biosynthesis, was isolated from hairy root cultures of Scutellaria viscidula Bunge by rapid amplification of cDNA ends (RACE). The full-length cDNA of S. viscidula CHS, designated as Svchs (GenBank accession no. EU386767), was 1649 bp with a 1170 bp open reading frame (ORF) that corresponded to a deduced protein of 390 amino acid residues, a calculated molecular mass of 42.56 kDa and a theoretical isoelectric point (pI) of 5.79. Multiple sequence alignments showed that SvCHS shared high homology with CHS from other plants. Functional analysis in silico indicated that SvCHS was a hydrophilic protein most likely associated with intermediate metabolism. The active sites of the malonyl-CoA binding motif, coumaroyl pocket and cyclization pocket in CHS of Medicago sativa were also found in SvCHS. Molecular modeling indicated that the secondary structure of SvCHS contained mainly α-helixes and random coils. Phylogenetic analysis showed that SvCHS was most closely related to CHS from Scutellaria baicalensis. In agreement with its function as an elicitor-responsive gene, the expression of Svchs was induced and coordinated by methyl jasmonate. To our knowledge, this is the first report to describe the isolation and expression of a gene from S. viscidula.
